Comparison of 5S and 45S rDNA genes of Crepidiastrum and related species, Youngia by fluorescence in situ hybridization in Korea.

Somatic metaphase chromosomes of Crepiastrum chelidoniifolium, C. denticulatum, C. lanceolatum, and C. sonchifolium were investigated by aceto-carmine staining and FISH using 5S and 45S ribosomal DNA genes. All species had commonly the chromosome number of 2n=2x=10 and most of the chromosomes submetacentric and subtelocentric. Physical mapping of the two multigene families (5s and 45s rDNA) was carried out by fluorescence in situ hybridization (FISH) using digoxigenin-11-dUTP-labelled 5S rDNA and biotin-16-dUTP labeled 45S rDNA as probe. In FISH, a pair of 5s rDNA signal was detected on the end of short arm of chromosome 4 in C. lanceolatum, C. chelidoniifolium and C. denticulatum but, C. sonchifolium showed different 5s rDNA site on the chromosome 3 around centromere. The 45s rDNA loci was observed on the secondary constriction in third chromosome as we guessed in C. sonchifolium, C. chelidoniifolium and C. denticulatum but, the other species, C. lanceolatum showed different loci on the chromosome 1 because secondary constriction in this species had in first chromosome. These cytological data of the four species show that different two rDNA genes are useful makers to characterize among the genus Crepidiastrum. The three patterns of two genes loci in this genus support that C. chelidoniifolium and C. denticulatum are cytotaxonomically the most similar to each other among the four Crepidiastrum species. But, C. lanceolatum and C. sonchifolium were different cytological relationships among this genus.
