Identifying North American sedges (Carex & Kobresia, Cyperaceae) via DNA barcodes: a regional approach. With more than 2000 species worldwide and greater than 480 species in the flora of North America, north of Mexico, the sedge tribe Cariceae is one of the most taxonomically diverse and ecologically significant clades on earth. Unfortunately, morphological species identification is difficult due to the tribe’s great taxonomic diversity, reduced morphology, and complex taxonomy, and thus sedge species are often ignored not only by the public, but even by scientists. In order to better understand Cariceae biology and to aid in the discovery and conservation of sedge taxa, we examine the utility of DNA barcodes for identifying species within tribe Cariceae. In a previous study involving five proposed chloroplast barcoding loci and nearly comprehensive sampling of three of the most taxonomically well-known sections in the tribe, our results suggested that ca. 60% of species would be resolvable, and that no multilocus barcode could significantly improve species resolution over the use of matK alone (i.e., 57% vs. 60%). Using matK and a significant increase in our taxonomic sample size, we re-examine this problem using a regional approach to DNA barcoding. Our results suggest that when barcoding queries are restricted to a particular region (e.g., a state or province), species resolution using matK can be >90% in some cases. Consequently, we suggest that the identification efficiency of current barcodes could be increased by a regional approach to DNA barcoding. Our results also suggest that matK is an informative phylogenetic marker. Phylogenetic relationships based on sequences from multiple individuals for most of the North American taxa, north of Mexico, will be discussed. Log in to add this item to your schedule
