Mapping 3D Xylem Networks In Vitis vinifera Using High Resolution Computed Tomography (HRCT).

Recent developments in High Resolution Computed Tomography (HRCT) have made it possible to obtain 3D information on xylem structure far more rapidly than by physical sectioning. HRCT is a nondestructive technique for visualizing the interior of solid objects and providing information about their 3D geometry, and current instruments are able to resolve features at the level of approximately 4.5 µm. Internode sections of Vitis vinifera L.‘Chardonnay’ were dried and imaged using HRCT at the ALS Synchrotron in Berkeley, CA. Image analysis revealed that the 3D vessel network could be resolved, allowing detailed information about vessel wall overlap and the location of pit membranes. Vessels greater than 20 µm in diameter are easily visualized, as well as the connectivity of the network. Previous vessel reconstruction methods involving serial sections have been useful, but are time consuming and labor intensive. This method allows for the rapid, detailed visualization of continuous vessel networks and provides the a new method for understanding xylem networks. Individual vessels, groups of neighboring vessels, or entire stems can be easily resolved with this new technology. Detailed data from the HRCT scans are now being integrated into mathematical xylem network models to more accurately predict hydraulic conductivity and simulate a variety of physiological scenarios resulting from the introduction of air emboli or pathogen infections.
